OxBS-seq discriminates between 5mC and 5hmC using chemical oxidation of 5hmC to 5-formylcytosine (5fC). Bisulfite treatment causes 5fC to be deformylated and deaminated to form uracil. Subsequent detection of 5hmC can be achieved by comparing BS-seq and oxBS-seq.
