Profiling miRNA expression in human stem cells using next-generation sequencing

Application of massively parallel sequencing to microRNA profiling and discovery in human embryonic stem cells.

New sequencing technologies such as Solexa/Illumina sequencing provide an ideal method for measuring the abundance of small RNA molecules in specialized cell types.  In this article in Genome Research, Ryan Morin describes novel techniques for analysis of Solexa/Illumina small RNA libraries and reveals the differences in the microRNA repertoire of human embryonic stem cells during differentiation.  This article also describes many previously undiscovered human microRNA genes, leading to the suggestion that many novel microRNAs will be discovered in the near future.
