Common regulators between autophagy and cell death
We developed an efficient flow cytometry and RNAi-based
method to identify genes that function in starvation-induced autophagy. We applied this method to 20 known cell death
genes in Drosophila, and identified 6 with an effect on autophagy. Of
particular importance, we are the first to associate an effector caspase Dcp-1
and IAP family protein Bruce with starvation-induced autophagy. Using Drosophila
mutant and transgenic lines, we further show that the caspase is required for
starvation-induced autophagy and is sufficient to induce autophagy in vivo.
This study provides new insights into the molecular mechanisms that
coordinately regulate autophagy and cell death in vivo.
